Thousands of cars on our dyno, the worst I have seen is a lifted head/nitrous explosion on a diesel puller pushing 105psi. Other than that, if the motor is assembled correctly you should not have a problem, the dyno will not abuse the motor any more than you would on the street with sticky tires.
If its a new motor braking in the engine on a dyno is better then breaking it in on the street.
If its not a new motor chances are quite slim it will break. A dyno just measures from RPM point 1 to RPM point 2 in gear clutch out 1 pull pedal to the floor. So unless the motor is already hurt you shouldn't worry much.

There was a motorcycle gathering down south that had several bikes blow up. They were having a contest to see who could get the most power down and they would over-rev and push the bikes to thier limits. So if you push it beyond it's limits (just like on the street), it can go. If you are just tuning and can see when the power starts to dive or know the RPM limit then I'd say you are safe.
