In a healthy engine, there is no vacuum in the line. It flows filtered/MAF measured air from in front of the TB blades, to the valve cover, where it is pulled through the engine by the PCV valve to eliminate harmful crankcase vapors. If for some reason the PCV valve can't handle the volume of crankcase vapor... the valve is plugged, or you are getting excessive ring blowby, pressure will build in the crankcase and force the vapors back through the valve cover/line to the TB, through the top of the TB, and they get sucked in through the throttle blades.
