I have a 2000 camaro ls1 A4 Lm2, y pipe, airlid. I want to get my car tune at this place in Jersey called Time Tune Performance. If anyone has ever gone there is it worth it. I believe its $400 dyno and tune.. Is it a big horsepower gain?

I'd wait until you get headers. Best thing you could do to that car is call Yank and buy a SS3600 torque converter.
I ran full exhaust, lid, and that converter for a year (3.23 car); it was an absolute blast.
And keep your transmission cool and you shouldn't have too much trouble, I'm running G5x2 cam, full bolt on, and a PT4000 (3.73 now w 315 drag radials) and still on the stock tranny. Bought it with 35,000 and been running a converter for 15,000 miles (55,000 now). Although if it's a DD I would refrain from the drag radials because my experience with the stock A4 may not be totally the norm...

TTP is a reputable shop I believe. They did a LOT of work for GM High Tech Performance. I would ask them if you got the tune now, and added some headers, intake, converter, etc later if they could touch up the tune for a small fee. If so, then it might be worth it. Most shops will touch up an existing tune for very little if you make small changes.
Of course if you do things like a supercharger, heads or larger cam then you would need another tune at full price.

I don't see yours putting down any more than 325, if you had a 2001 or 2002 with the ls6 intake then yes you could be higher. Is his a 01 or 02 if so he has the ls6 intake. And if he has a M6 it will lose less HP from the crank to the rear tires.

I don't really see a point in getting a dyno tune until you have all bolt ons and a cam.
You can look into getting a mail order bolt on tune from Frost or somebody like that. I think Frost still charges $125 but you might want to confirm that on LS1tech. Your car will be down for a few days though. I found somebody local that did my bolt on tune for a good price and I could feel the difference and I even get better mpg now.

There isn't much to be gained with a tune on a stock setup. The timing is already agressive on a stock tune. However, the stock calibration is a little rich at WOT and there are a few hp to be potentially gained by taking a little fuel out. Not worth $400 IMHO and a reputable shop will tell you that. OTOH, even a seemingly good running engine may make less hp when tested, pointoing towards a previously unsuspected problem. But generally speaking, it isn't worth it to tune a stock (or nearly stock) vehicle.
