Flexplate for LT1/th400

Will the stock LT1 153 tooth flexplate work with a th400? If not what is required?

Yes, if the bolt pattern of the torque converter is designed for the 153 tooth flywheel.

With your setup, you might want to give serious consideration to an SFI-rated aftermarket flexplate.
