You actually can. but you need a special tool to do it. You need an adapter that fits on where you put the oil filter, and then you run pressurized oil into it. Never done it before, but thats what ive read in the 'how to build lt1/lt4 engines' book.
If this is a new motor and you havent primed it at all, your going to have to remove the intake and actually spin the pump. The above way is only if the engine has been sitting for a couple weeks or more fully assembled, but hasnt been started yet (but has already been primed once before with the pump).

is it in the car or on a stand.
if its on a stand flip the motor over and fill the sump.
fill the oil filter up. put the oil pan on.
when its back in the car disconnect the coil wire and turn over the motor till you get oil pressure. or just take the intake off.
if its on a stand flip the motor over and fill the sump.

take off the intake, fuel rails egr tube ect. take the bolts that hold it on.
if its running why do you need to take it off? are you getting no oil pressure?
if you have no experience doing this then you should not do it. it will be more of a mess than what you are expecting.

If it's running and doesn't have any oil pressure, priming it is not your problem. Priming is to prevent a dry start in the system.. the system will eventually get pressure with the engine turning.
