Physically? It should. It was designed to fit anywhere the 4L60 fits. My understanding is that you shouldn't even need to change the driveshaft length.

I've been waiting for someone to put an LS3/6L80 combo in a 4th gen.

See, that's what I thought.

http://www.6l80.com/ guys tell me no. They say it is physically much bigger so I don't know.

I'm looking to do an LS2/LS3/L92/L99 swap with a 6L80E.

They said it wouldn't fit in the transmission tunnel. I dunno.

It doesnt look any bigger than my 4L80e, matter of fact a little more narrow, which was the only thing that was remotely a problem. If you can find a comparison pic between it and the 4L80e it should answer most of your questions. It didnt take much at all to get my 4L80 in the tunnel, so I dont think it should be a problem. If it fits in a Vette it'll fit in an Fbody.

Just saw the picks of it compared to a 4L65e, no problem with that thing fitting my 4L80 is huge compared to a 4L60.

No one has been able to put one in a Trailblazer yet, so with all of that room, I find it hard to believe that it would be an "easy" swap into an F-body.

Looking at the tail shaft I'm guessing you would have to "massage" the tunnel a bit.
