I've been to almost every "nice" restaurant in the city and here are my recommendations:

If you want something up-beat and entertaining:
Kobe
Elixir

Steak:
Gotham
Hys

Romantic:
Villa del Lupo
Cafe de Medici

West Coast:
La Terraza
Ballentynes

Seafood:
Joe Fortes
Aqua Riva

They're all good and you would need to make reservations.

I voted for Cloud 9 on Robson. A little more formal but you don't have to wear a suit. Awesome view for out of towners revolving around. I little pricey too but great food and service. Everyone should experience this place at least once. Last time I was there I had tenderloin steak and shelled king crab with bernaisse sauce. I feel like making a reservation just thinking about it.

If you are willing to go about 5min out of town you can go to the BEST restaurant in Vancouver...Lumiere. Done, case closed (I work there ) If you want to pay alittle less, go next door to Feenies. Both are owned by Rob Feenie, who has a cooking showon the Cooking Network, Wed @ 5:30 .

If you want the best Japanese food in town then go to where all the movie stars go, TOJO's on Broadway.
