Took out my lifters this weekend (stock 94 LT1), and they each have a hole on one side only. These holes are positioned to face each other for each cylinder.

What are these for, and do they have to face each other?

I beleive they are there for oil flow to block lifter walls. I don't recall the oem lifter set-up, but I don't believe the rotation in the slot matters as long as the retaining plates fit when the lifter is seated in place.

On lifters with a link bar (non-LT1), one places the link inward. But as I mentioned above I am not familar with any requirement that the LT1 hyd-rollers be positioned in the bores in special fashion other than that fit well beneath the retaining plates. I'm no professional engine builder so I don't want to say I am 100% certain.

They do not rotate in their bores once you install them. The roller will only ride on the cam if it is parrallel to it. If it turned you would have major cam damage.

It's gonna be kinda hard for them to rotate with the lifter guides on there that keep them from rotating. They cannot rotate because of the roller that is on the cam end of the lifter. Regular SBC non-roller ones are free to rotate.

Doesn't matter, anyway. The hole is there to pass oil into the lifter. That's how the lifter gets it's oil and is able to pump it up through the pushrod to the rocker arm.

I thought there were some type of kits out there that were made specificly to keep them from rotating, only reason i posted that, I apolgize for the bad info.
